Looking at glass test tubes on Amazon, I see that they come with either cork or rubber stoppers. Is one of these better than the other? Also, will glass ones with stoppers leak and need to be kept upright? I planned on getting rounded-bottom tubes because they look easier to dig errant seeds out of the bottom of, but if it has to be upright, maybe flat-bottomed is better?

No difference observed between cork and rubber stoppers.
I tried some cheapcheap plastic tubes with even cheaper stoppers and they leak if horizontal.
I usually use a test tube rack to keep them vertical, just to be safe.
