Did you put it in the plastic dome and in the sun before or after you got the dark spot. It looks to me like that dome may have acted like a magnifying glass and fried the leaf.

Was that in direct sunlight? I think that was the problem. It should recover fine, but keep it out of direct sun. Neps don't need that much light.

I think that it should be. The growth point may die off. It depends on how damaged it is. If it does, it will sprout a new one at a leaf junction or put out a basal.
