if you put it upside down will all the water fall out and the bugs? and will it die?

I don't know if it'll die, but all the 'water' and bugs will fall out of the pitcher.
It does require a lot of energy to make digestive fluids, so I'm pretty sure the plant won't like to lose it

are you supposed to put water in the Nep traps? or do they make their own fluid?

They make their own fluid. The only time I tried putting water into Nep traps was when I first got my N. sanguinea in the mail and all of the fluid was out of the traps. I added water back into them and the traps all promptly turned brown and died. It could have just been due to the shock from being mailed, but since then I've not tried to add water to my Nep traps.

It's strongly advised not to put water in the nep pitchers because the digestive fluid in the pitchers should not be diluted. That's one of the reason why the neps have the lids, to keep rain water out of the pitchers. I'm not sure about adding water could be the reason for the pitcher to die, but if it's pH sensitive, then adding water might kill that pitcher.
