Hi,
After many years i am back here and i have a very interesting project:

I am very successful in culture of U. menziesii. And now i will try it in tc!

I have harvested some fresh seeds! Can anyone tell me a sterilize protocol for this seeds? I think they are soft shelled like other utric seeds.

And whats the best media for this tuberous Utric? 1/6 MS, 30g sugar, pH 5.7? Could it be work?
I think the sugar level must be high for new tubers!

What do you think?

Best regards from germany
Tobi

Hey Tobi,

Did you end up getting seed into culture?
I would try 10% bleach for 4-5 minutes then rinse. If you want to neutralize the free chlorine, you can then rinse with sterile water with filter-sterilized 1.2g/L ascorbic acid. This helps minimize damage from any bleach remaining on the tissues.

Your MS is low but I'm sure would still be acceptable. 1/2 or 1/3 MS should also be fine.

Take this advice with a grain of salt since I haven't worked on this species before.
